A scanning UV-Visible spectrophotometer is a type of spectrophotometer that measures the ratio of light passing through a sample across a range of wavelengths in the ultraviolet and visible spectrum. This allows it to capture a complete absorption spectrum, revealing how a sample interacts with light at each individual wavelength ; A spectrophotometer operates on the principle that molecules absorb light at specific wavelengths. It measures this by comparing the intensity of light before and after it passes through a sample .

 

The fundamental relationship is described by the Beer-Lambert Law, which states that the amount of light absorbed is directly proportional to the concentration of the absorbing substance and the path length the light travels through the sample . This is the basis for quantitative analysis, allowing you to calculate an unknown concentration if you know the substance’s specific absorption characteristics. To capture a “scan,” the instrument systematically varies the wavelength of light over a defined range. The result is a graph, or spectrum, plotting absorbance (or transmittance) against wavelength. The wavelength where a sample absorbs the most light is known as λmax, a key characteristic used for identification and analysis .

 

A Scanning UV-Visible (UV-Vis) Spectrophotometer is the workhorse of the modern analytical lab. Unlike a fixed-wavelength photometer, a scanning model can sweep across a range of wavelengths—typically from 190 nm to 1100 nm—to produce a full absorption spectrum.
Think of it as the difference between taking a still photo (fixed wavelength) and recording a high-definition video (scanning) of how a substance interacts with light.

Instrument Design : Single Beam vs Double Beam
Scanning spectrophotometers come in two main optical configurations, each with its own workflow

–> Single Beam : This design has a single light path. To use it, you first measure a “blank” (the solvent without your sample) to establish a baseline. Then, you replace the blank with your sample and measure its absorbance. It’s a simple, robust, and often more affordable design, ideal for routine measurements where the baseline is stable .

–> Double Beam : This design splits the light into two paths: one for the sample and one for a reference (the blank). The instrument continuously compares the two beams, which effectively cancels out any fluctuations in light source intensity or drift in the detector. This results in excellent stability, higher accuracy, and faster scanning, making it the preferred choice for research and demanding applications
